Malaria is a mosquito-borne infectious disease, transmitted to human by an infected female Anopheles mosquito. Malaria is usually confirmed by the microscopic examination of blood smears or by antigen-based rapid diagnostic tests (RDT).
A parasite level in the blood of greater than 100,000 per microliter in low-intensity transmission areas or 250,000 per microliter in high-intensity transmission areas can be diagnosed as malaria. The accuracy of blood smears ranges from 75 to 90% in optimum conditions to as low as 50%. RDT, by contrast, are often more accurate at predicting the presence of malaria parasites. But they are widely variable in diagnostic accuracy and specificity depending on the manufacturer, and are unable to tell how many parasites are present. In order to increase the analytical accuracy and quantification across a wider range of parasite densities, in the past decade, PCR for malaria have been developed.
